The Jolly Grant airport is only about 40 km from Dehradun, so a cab from there will get you to the Yamunotri route in roughly an hour before the long stretch. If you prefer to break up the drive, staying a night at Prakriti The Retreat in Harsil puts you only 32 km from the temple, making the final leg a quick auto or brisk walk. This can help you avoid the peak afternoon traffic on the Dehradun Yamunotri road.
